Abstract

 Process Hazard Analysis (PHA) is a critical aspect of ensuring safety during the scale-up of bioprocessing systems. Scaling up from bench to industrial scale introduces complexities in design, operation, and safety that can lead to unforeseen hazards if not addressed properly. This paper investigates the application of PHA techniques in identifying and mitigating risks during bioprocess scale-up. A systematic methodology combining Hazard and Operability (HAZOP) analysis, Failure Mode and Effects Analysis (FMEA), and quantitative risk assessment (QRA) is proposed and tested in a case study involving microbial fermentation. The findings highlight the importance of integrating safety analysis into the design and operational phases to minimize risks, reduce downtime, and ensure regulatory compliance.
